If killing the original Cultist kills every cult member, the Cult has a balanced weakness.
If Serial Killer can be Culted and still retain his role, things can get unbalanced.
If Mafia can be Culted, Cult can be unbalanced. If Culting Mafia kills the Cultist, he won't be too overbalanced.
Most of the rest depends on how strong your town is. Some games have special cops who look for the serial killer or Cultists. You can look into a Priest role -- someone who passively affects villians who visit him during the night. He can often turn Serial Killer into a Survivor (resistance to a lynch, win if they survive to the end) and is generally immune to being Culted.
If you make a game with powerful 3rd parties, it can be balanced -- the 3rd parties will cancel each other out. Your game will have more enemies than mafia, though, so you should create your town with that in mind.
