A PS1 Game, no. Not unless you buy it off the PSN

Unlike PS2 Support, PSX support is in all PS3s.
My friend put Metal Gear Solid in a 80gb PS3 (without PS2 support) and it worked.
You can also buy PSOne games off PSN and play them on both the PS3 and PSP, but unless you want psp support, there is no use buying them again.
