The Fighting Misty

I can see Giovanni attending this battle facility in the anime; he's still a Gym Leader. He could go on an undercover mission, or use this facility as an excuse for his ventures in Unova. ("Hey Giovanni, isn't it funny how Team Rocket appears in Unova at the same time you do?" "Naw, I came here for Cynthia's little project.")

Last but not least, I won't mention her by name, but a certain red-headed character probably appears in the new games too.

He didn't say the name that starts with an upside-down W(I think it's May, she had a red bandanna) because then people would count how many times he said it.

She was acting as temporary Gym Leader.

I doubt Giovanni is still a Gym leader in the anime.

I'm theorizing that Giovanni may be teaming up with Ghetsis in BW2 because Gio is always seen as the series' primary villain.

As for Misty, I would be surprised if her appearance in the game would give her a ticket to appear in SSB4 as BW2's rep while still being associated with Gen 1 (Just like Red representing the Gen 3 games with his FR/LG outfit despite being Gen 1 material) because it's becoming a pattern that only one Pokemon rep in SSB is a current gen Pokemon while everyone else is from the first gen (Pichu was the only second gen rep in Melee, and Lucario was the only fourth gen rep in Brawl).

I seriously doubt that any Gym Leader would be a playable character in SSB4.

I don't see why not. When he's too busy being evil to battle in the Gym, he has people cover for him. And a Gym would be a great place to test out his strong Pokemon. (like with Mewtwo.) I don't really see a reason why he can't still be a Gym Leader.

Giovanni and Ghetsis teaming up makes no sense.

And Misty, heck, no gym leader but especially not Misty, will ever be a PC in SSB.

Perhaps he's under investigation in Kanto and had to duck out for a while?

The episode where Giovanni uses a Mewtwo on Gary is the greatest Pokemon episode of all time.

People can make fun of Misty and Brock all they wont, being sad day when original and great characters serve nothing more as material to make jokes about it. But if they do appear or return in anime thanks to their appearance in upcoming games, i know i m going to enjoy in it being interested in seeing what may have changed in mean time. Especially about Misty since we haven't see her since 2005.

Writers left some things open ended about his doctor career providing them with options to have him show up, and Misty story was still left unresolved,. Like her goal of water master with writers not defining what she plans to do in future, given how she only took gym over out of necessity(used just as vector to have her leave).

That being said i would like to see cook showdown between Cilan and Brock. As for Misty coming for some water tournament and show new pokemon would be pretty great too. Or helping Iris about potential gym issues having quite few things in common.

Also several older gym leaders appearing doesn't change anything, because if writers decide to affect anime by this, leaders which will get opportunity to shin would be ex main characters.
