In all honesty, aside from Project Café, the one announcement I'm looking forward to most is anything regarding Pikmin 3. Seriously, this has to be one of the most under hyped series of all time. Sure, it's not particularly flashy or adventurous, but this series pioneered the GameCube and made it what it is today, and if Café turns out to be a true sequel to the 'Cube, I'd love to hear that Pikmin 3 is a launch title. Still, based on previous interviews, it sounds like Pikmin 3 will end up being on the Wii. I'm really just waiting for them to show us what's in store. Who wants to join me in crossing fingers for a trailer..?
Aside from that, a new Animal Crossing game would be nice, and it should be no secret how in love with the Paper Mario series I am, so I'd love to see some in-game footage or a trailer from the 3DS installment (though I highly doubt that will happen). I'm really not all that excited about Skyward Sword, and I'm growing tired of all of this motion control stuff, so I don't really care about that unless they reveal something new the game will actually bring to the series. A new Mario platformer is in order, I believe, though, again, I'm not keeping my hopes up.
I have a hard time believing Nintendo's spotlight will actually be able to compare to Sony's. Sure, Sony had a terrible year, what with the earthquake and the network hack, but with titles like Resistance 3, Uncharted 3, FarCry 3, and all that beautiful new hardware like the new PSVista (which looks friggin' incredible) launching with LittleBigPlanet Touch, I think Sony is the E3 king this year. Though, I doubt anyone on this forum will ever admit to that. Hell, I'm just as excited as anyone to see Nintendo surprise us this year, so I'm not skeptical. I just haven't been impressed by Nintendo for a while. So, there's your challenge, Nintendo. Impress me!
